On Thursday 24 September at 13:00, Amalie Lykke Olsen defends her PhD dissertation entitled “Lipid composition shapes the efficiency of LNP-mediated CRISPR delivery”.

CRISPR is a genome-editing technology that enables targeted changes to DNA and therefore has the potential to treat diseases caused by genetic alterations. However, realising this potential requires the safe and efficient delivery of the necessary CRISPR components to the tissues and cells where the genome editing is intended to take place. Lipid nanoparticles, small lipid-based particles capable of encapsulating and transporting molecules such as RNA, have emerged as a promising alternative to traditional delivery methods. However, their delivery efficiency depends, among other factors, on the types of lipids that are used and their relative ratios. Furthermore, different cell types may respond differently to the same nanoparticle formulation. A better understanding of how lipid nanoparticle composition affects the delivery of CRISPR components to different cell types is therefore needed.

A new PhD study from Aarhus University addresses this question by systematically varying the composition of lipid nanoparticles and investigating their properties and ability to deliver RNA and CRISPR components in different cell models. The project thereby contributes to a better understanding of how lipid nanoparticles can be designed to enable efficient genome editing.
