On Friday 2 October at 14:00, Mie Kristine Just Pedersen defends her PhD dissertation entitled “Neuropathological studies of alpha-synuclein propagation and heterogeneity in Lewy body disease using archival tissue from the Danish Brain Collection”.
Examination of the brains of deceased persons provide important insights to diseases of the brain. Lewy body disease (Parkinson's disease, Parkinson's disease dementia, and dementia with Lewy bodies) is characterised by the build-up of the protein alpha-synuclein. This protein can be visualised by immunohistochemical techniques. One of the world's largest brain collections are located in Denmark, namely the Danish Brain Collection. The brain tissue has been stored for more than 40 years. Therefore, the first part of the PhD project was to demonstrate that the archival brain tissue remain suitable for immunohistochemical analyses of alpha-synuclein. The results show that alpha-synuclein is still detecable. The following studies investigates the propagation of alpha-synuclein. The main findings are that anatomical connections are important for the accumulation of alpha-synuclein in a given region, and that distinct propagation patterns can be identified in Lewy body disease. The project thereby contributes to our understading of the pathology in Lewy body disease, as well as establishing the utility of the Danish Brain Collection as a resource for scientific research.
